Review highlights recent advances in chiral indole-fused ring synthesis, indicating future directions for innovation.
Chiral indole‐fused medium and large rings are key structural motifs prevalent in a diverse array of natural products, chiral catalysts, and pharmaceutically active compounds. Despite their significance, the catalytic enantioselective construction of these ring systems remains a formidable and underexplored challenge. In response, chemists have recently devised innovative strategies that are propelling the rapid emergence of this field, affording novel scaffolds with promising applications in medicinal chemistry and asymmetric catalysis. This review provides a concise yet comprehensive overview of the latest advances in the catalytic enantioselective synthesis of chiral indole‐fused medium and large rings. Emphasis is placed on the underlying synthetic strategies, reaction mechanisms, catalytic models, and the potential applications of the resulting products. Furthermore, an outlook on the remaining challenges and future directions is presented to inspire continued innovation in this evolving field and related disciplines.
