Study evaluated a new drug delivery system for better nifedipine release, suggesting improved patient compliance.
Abstract:Nifedipine is a calcium channel blocker widely used in the management of hypertension andangina pectoris. However, its short biological half-life and limited bioavailability necessitatefrequent dosing, which may reduce patient compliance. The present study aimed to develop andevaluate a novel gastroretentive floating matrix drug delivery system for the sustained release of nifedipine. Floating matrix tablets were prepared using suitable hydrophilic polymers and gas-generating agents by direct compression method. The formulations were evaluated for pre-compression parameters, post-compression characteristics, buoyancy properties, drug content uniformity, swelling behavior, and in vitro drug release. The prepared formulations exhibitedsatisfactory physicochemical properties, acceptable floating lag time, and prolonged floatingduration. In vitro dissolution studies demonstrated sustained drug release over an extendedperiod, following diffusion-controlled kinetics. The optimized formulation showed desirablefloating characteristics and provided controlled release of nifedipine, which may enhance gastricresidence time, improve bioavailability, reduce dosing frequency, and increase patientcompliance.
