Review discusses quality assurance applications of Process Analytical Technology in pharmaceutical manufacturing, indicating significant improvements.
Conventional pharmaceutical production has largely depended on predefined manufacturing instructions followed by laboratory examination of the finished product. Although this approach can confirm compliance of tested samples, it provides limited information about process behaviour while manufacturing is in progress. Process Analytical Technology (PAT) changes this perspective by introducing timely measurements, multivariate analysis, and process-control strategies directly into production. The concept, formally encouraged through the U.S. FDA PAT framework, complements the Quality by Design principles described in ICH Q8(R2), quality risk management in ICH Q9, and the pharmaceutical quality system in ICH Q10. This review discusses the scientific basis, regulatory framework, principal tools, and quality-assurance applications of PAT. Particular attention is given to near-infrared and Raman spectroscopy, chemometrics, real-time process monitoring, and real-time release testing. Applications during material identification, blending, granulation, drying, compression, coating, continuous manufacturing, and bioprocessing are reviewed. Practical benefits and implementation difficulties are also considered, together with emerging developments involving machine learning, data fusion, digital twins, and Industry 4.0. Overall, PAT supports a preventive and knowledge-based quality system in which manufacturing quality is monitored and controlled during processing rather than assessed only after batch completion.
