The phosphorylation of bovine liver nucleoside diphosphate kinase (EC 2.7.4.6) by AT32P and the dephosphorylation of 32P-labeled enzyme by dGDP were studied by a rapid mixing technique. When investigated in the presence of Mg++, the rate of the phosphorylation and the dephosphorylation reaction was found to exceed that of the over-all reaction, indicating that the phosphorylenzyme is an intermediate in the nucleoside diphosphate kinase reaction. Furthermore, kinetic data consistent with a ping-pong mechanism, according to Cleland's terminology, were obtained.
