The methodology is described, as developed at the Albany NIH Biotechnological High Voltage Electron Microscope Resource, for the three‐dimensional reconstruction of objects in thick sections. The reconstructions are obtained from projection sets which are recorded by high voltage electron microscopy. The different steps of the procedure are illustrated in detail, using the cilium reconstruction as an example.
