In multi-enzyme complexes, different enzymes are associated non-covalently and catalyze sequential steps of biochemical pathways. One of the interesting aspects of complexes having a high degree of organization, such as the α-keto acid dehydrogenases or fatty acid synthetase (cf. Reed and Cox, 1966; Lynen, 1964), concerns their biosynthesis. We have been studying the synthesis of the E. coli K12 pyruvate dehydrogenase complex and its genetic control (Henning, Herz, and Szolyvay, 1964a).
