An isotope derivative method for determination of inorganic pyrophosphate (PP i ) in biological material in amounts down to 10 pmoles is described. The method is based upon measurements of radioactivity in [5‐ 3 H]UTP formed in UDP‐glucose (UDPG) pyrophosphorylase reaction using a highly purified preparation of UDPG pyrophosphorylase from calf liver and [5‐ 3 H]UDPG. The optimal conditions for reaction between [5‐ 3 H]UDPG and PP i was brought about by addition of phosphoglucomutase, NADP + and glucose‐6‐phosphate dehydrogenase. The method was specific and no influence from endogenous metabolites could be demonstrated. By this method the total concentration of PP i in rat liver and guinea pig liver was found to be in the range from 0.5 mM to 1.4 mM.
