To determine the differential roles of central target ablation versus peripheral deafferentation on neurogenesis, neuronal maturation, and the expression of B-50/GAP43 and olfactory marker protein in the olfactory neuroepithelium.
Induced olfactory pathway lesions through either peripheral chemical deafferentation using Triton X-100 or central olfactory bulbectomy.
Tracked neuroepithelial regeneration and neuronal differentiation using mRNA probes and antibodies directed against B-50/GAP43 and olfactory marker protein (OMP) for up to 3 months post-lesion.
Both peripheral deafferentation and bulbectomy initially induced a first stage of regeneration marked by the emergence of immature olfactory receptor neurons expressing high levels of B-50/GAP43.
A second stage characterized by B-50/GAP43 downregulation and full OMP expression occurred exclusively following Triton X-100 lesion where the olfactory bulb remained intact.
Target ablation via bulbectomy resulted in persistent B-50/GAP43 expression up to 3 months post-injury and a failure to reestablish a full complement of mature OMP-expressing neurons.
