To precisely define and physically map the type 1 diabetes susceptibility interval (IDDM12) within chromosome 2q33 near candidate genes CTLA4 and CD28.
Genotyped 10 polymorphic markers across a ~300-kb genomic interval in a multiethnic cohort of 178 simplex and 350 multiplex families (U.S. Caucasian, Mexican-American, French, Spanish, Korean, and Chinese).

Evaluated genetic association and linkage using transmission disequilibrium test (TDT) and Tsp analyses.
Transmission disequilibrium testing revealed significant linkage and association with three polymorphic markers within CTLA4 and two flanking markers (D2S72 and D2S105), but not with distant markers including candidate gene CD28.
Tsp analysis demonstrated that significant association was strictly confined to the three polymorphic markers within the CTLA4 gene itself.
Disease-linked markers localized within a single 100-kb phagemid artificial chromosome (PAC) clone, indicating IDDM12 is either CTLA4 or an adjacent gene.
