Liver perfusion in Squalus suckleyi demonstrated hepatic ketone production and hormone-mediated hemodynamic changes, but perfusion with insulin and glucagon did not alter glucose balance.
First description of a viable liver perfusion model in a shark species, demonstrating hepatic ketone production but no changes in glucose balance with insulin/glucagon.
Viable shark liver perfusion model established; extends comparative ketogenesis research but leaves open mammalian translation.
We describe a viable liver perfusion in a shark species for the first time as determined by oxygen consumption and hormone-mediated changes in hemodynamics (angiotensin II, ANG II). In addition, removal of major energy metabolites confirms hepatic ketone [3-hydroxybutyrate (3-HB)] production by an elasmobranch liver. Perfusion with heterologous insulin and homologous glucagon did not cause changes in glucose balance, however, possibly demonstrating differences in glucose metabolism in this taxon as compared with more derived vertebrates.

Schoen et al. (2023) studied this question. Liver perfusion with heterologous insulin and homologous glucagon was evaluated on Glucose balance, oxygen consumption, and hormone-mediated changes in hemodynamics. Liver perfusion in Squalus suckleyi demonstrated hepatic ketone production and hormone-mediated hemodynamic changes, but perfusion with insulin and glucagon did not alter glucose balance.
