Reveals microbial community composition in strawberry rhizosphere soil, suggesting insights for cultivation strategies.
16S rRNA and ITS amplicon sequencing performed on strawberry rhizosphere soil microorganisms. In this study, rhizosphere soil samples attached to the root surface of cultivated strawberry plants were collected in the field under standardized sampling specifications. We extracted total microbial genomic DNA from each soil sample with commercial DNA extraction kits, detected DNA concentration and purity through agarose gel electrophoresis and ultraviolet spectrophotometry to ensure qualified DNA quality for library construction. Targeted amplification was conducted on the V3 and V4 hypervariable region of bacterial 16S rRNA gene and the ITS1 region of fungal internal transcribed spacer respectively. After PCR product purification, quantification and homogenization, high throughput sequencing was carried out on the Illumina sequencing platform. Raw sequencing reads will be deposited into public omics databases for permanent open access; subsequent bioinformatic analyses including sequence quality trimming, operational taxonomic unit clustering, species annotation, alpha and beta diversity calculation, and differential species screening will be implemented to reveal the composition, abundance and interaction patterns of bacterial and fungal communities in strawberry rhizosphere soil. The publicly available raw sequencing data can be reused by peer researchers for meta analysis of crop rhizosphere microbiomes, mechanism exploration of plantmicrobe interaction and screening of beneficial microbial strains for strawberry cultivation.
