Radioactive sedoheptulose 7-phosphate was shown to label specifically the l-glycero-d-mannoheptose residues of the lipopolysaccharide of a transketolase-negative mutant of Salmonella typhimurium. Phosphoheptose isomerase, the first enzyme in the proposed pathway of heptose biosynthesis, was shown to catalyze the reaction: sedoheptulose 7-phosphate → d-glycero-d-mannoheptose 7-phosphate. This enzyme was partially purified and utilized for the enzymatic synthesis of d-glycero-d-manno[14C]heptose 7-phosphate from [14C]sedoheptulose 7-phosphate. These findings support the proposed pathway for the conversion of sedoheptulose 7-phosphate to l-glycero-d-mannoheptose (Eidels, L., and Osborn, M. J. (1971) Proc. Nat. Acad. Sci. U. S. A. 68, 1673–1677).
