Cardiovascular performance was tracked by measuring coronary flow rate, left ventricular developed pressure, dP/dTmax, dP/dTmin, and heart rate.
Myocardial tissue was freeze-clamped at the end of perfusion to quantify calcium levels and tissue metabolites.
Perfusion with 500 mg/l MPSS produced significant inotropic stimulation and vasodilation, increasing coronary flow rate (+17% ± 4%), left ventricular developed pressure (+6% ± 1%), dP/dTmax (+13% ± 2%), and dP/dTmin (+9% ± 3%), while heart rate decreased (-14% ± 2%; all P < 0.05).
Perfusion with 100 mg/l MPSS had no detectable effect on coronary flow or contractility, whereas 1000 mg/l MPSS caused transient vasodilation followed by progressive myocardial depression.
The myocardial adenine nucleotide pool was significantly increased by 100 and 500 mg/l MPSS, while myocardial lactate content was elevated across all tested concentrations.
