To evaluate the potential role and therapeutic efficacy of PI3-kinase inhibitors in mitigating delayed cerebral vasospasm following subarachnoid hemorrhage.
Assigned 24 dogs with experimental subarachnoid hemorrhage induced by cisterna magna blood injections on days 0 and 2 to receive vehicle (DMSO), wortmannin, or LY294002 daily on days 2 through 6.
Evaluated basilar artery narrowing on day 7 via angiography and measured arterial morphology, Western blot protein expression, and PI3-kinase enzymatic activity.
Vehicle-treated control dogs developed severe arterial narrowing on day 7, showing a basilar artery residual diameter of 47.8 ± 0.8% relative to day 0.
Wortmannin and LY294002 successfully abolished PI3-kinase enzyme activity compared with vehicle, but neither drug significantly altered residual basilar artery diameter on day 7 or attenuated PI3-kinase protein expression (P>0.05, ANOVA).
