Overview

Tamoxifen may attenuate renal damage in hypertensive rats; leaves open translation to human hypertensive nephropathy.

Structured PICO

Does tamoxifen provide renoprotective effects and reduce inflammation and fibrosis in a rat model of DOCA-salt hypertensive nephropathy?

P
Population
22 male Sprague-Dawley rats (weighing 180 to 200 g) post-unilateral nephrectomy. 16 were implanted with deoxycorticosterone acetate (DOCA) strips (200 mg/kg) to induce hypertensive nephropathy, and 6 served as non-implanted controls.
I
Intervention
Tamoxifen (10 mg/kg) administered through gavage feeding, co-treated with DOCA strips for 2 weeks.
C
Comparator
DOCA strips alone without tamoxifen (n=8) and non-implanted controls (n=6).
O
Outcome
Systolic blood pressure (SBP) measured by tail-cuff method and protein expression of transforming growth factor-beta (TGF-beta), Smad, alpha-smooth muscle actin (alpha-SMA), E-cadherin, ED-1, and cyclooxygenase-2 at 2 weeks.surrogate
