Intramolecular resonance energy transfer probes, coumarin phosphodiester-linked fluoresceins (such as CPF2), were designed and synthesized. In a probe for ratiometric measurements, it is important to observe the emission of the acceptor, but self-quenching occurs in aqueous solutions. We show that the emission of the acceptor can be observed if a linker with low flexibility is introduced between the fluorophores.
