Harvested chylomicra from the thoracic ducts of olive oil-fed rats and washed them to eliminate soluble lymph proteins.
Fixed specimens in osmium tetroxide, embedded them in Araldite resin, and examined ultrathin sections using electron microscopy.
Chylomicra displayed circular cross-sections with diameters ranging between 100 and 3,500 mµ and showed uniform electron density throughout the interior.
A darkly staining peripheral boundary corresponding to minor lipids and proteins was identified as an interrupted pattern of minute discrete specks rather than a continuous film.
