Chemical synthesis study demonstrates photocatalytic cascade cyclization using BTFB for N-heterocycle formation, indicating broad utility in late-stage drug functionalization.
A photoredox strategy for synthesizing diverse polyfluoroalkyl-substituted N-heterocycles has been reported. Through in situ halogen exchange for activation of polyfluoroalkyl bromides, a photocatalytic cascade cyclization using 4-bromo-3,3,4,4-tetrafluorobutene-1 (BTFB) as the polyfluoroalkyl radical source is described for the first time. It introduces polyfluoroalkyl and terminal alkenes into N-heterocycles in a single operation without metal catalysts or redox reagents, featuring broad substrate scope and late-stage functionalization applicability.
