The study of plasmid DNA replication not only has yielded basic information on the mechanisms involved in the maintenance of these elements in bacteria, but also has led to the development of several model systems that have provided insights into the biochemical processes and genetic regulation of DNA replication in general. The various plasmid systems studied to date show that replicons vary considerably in their modes of replication, at least with respect to copy number and directionality of replication (Helinski 1976). A clear requirement in each case, however, is a mechanism of regulation of replication that ensures inheritance of at least one copy of the plasmid by each of two daughter cells after cell division.
