The RNA polymerase activity of the purified nuclei from the prostate of castrated rats was measured shortly after the subcutaneous injection of testosterone propionate to the animals. A significant enhancement of RNA polymerase activity was demonstrable within a few hours after the single injection of the androgen if the enzyme activity was measured in a reaction mixture of a relatively low ionic strength. Under the same conditions, RNA polymerase activity of thymus or liver nuclei of orchiectomized rats was not increased by the administration of testosterone to the animals.
