The experiments we describe here were undertaken to learn whether successive replications of the E. coli chromosome begin at the same site, and to study certain other aspects of the pattern of replication of the bacterial chromosome during steady exponential growth. Our approach has been to investigate the distribution of time intervals that elapse between the synthesis and the replication of a short segment of the bacterial chromosome. The procedure we have adopted for determining this distribution is as follows.
