To identify the molecular determinants that govern the distinct subcellular localization and trafficking of the two Hck tyrosine kinase isoforms, p59Hck and p61Hck.
Generated green fluorescent protein (GFP) fusion constructs of full-length p61(Hck), p59(Hck), their isolated N-terminal domains, and a palmitoylation-site mutant of p59(Hck).
Expressed the constructs in HeLa and CHO cell models to determine intracellular distribution using fluorescence microscopy.
Wild-type p61(Hck) localized selectively to lysosomal vesicles, whereas dual-acylated p59(Hck) localized predominantly to the plasma membrane with minor lysosomal association.
Isolated N-terminal domains were sufficient to target GFP to the respective subcellular compartments of each isoform.
Mutating the palmitoylation site of p59(Hck) shifted its localization to lysosomes, while both isoforms and the mutant remained detectable in the Golgi apparatus.
