The identification of a novel NKX2-5 mutation expands the known genetic basis for familial atrial septal defects and associated arrhythmias.
Highlights the need for genetic screening in familial atrial septal defects with conduction abnormalities; leaves.
Secundum atrial septal defects (ASDs) account for approximately 10% of congenital cardiac malformations.1 Most ASDs occur sporadically; however, familial forms have been described and are enriched for pathogenic variants in genes involved in cardiac development. Among these, NKX2-5, a cardiac-specific homeobox transcription factor essential for atrial septation and conduction system formation, is the most consistently implicated gene.2 Pathogenic variants in NKX2-5 are identified in a small proportion of sporadic ASDs and in approximately 8%–12% of familial cases.
