Resonance energy transfer (RET) is exploited in a novel assay of endoglucanase activity (see scheme below). The variation in the fluorescence intensity of a fluorogenic oligosaccharide is followed during its hydrolysis, allowing the determination of α-amylase activity, one of the starch-degrading enzymes, which is important for diagnosing diseases like parotitis (mumps) and acute pancreatitis. = sugar unit; F1,F2 = fluorophore.
