An enzyme has been found in soluble extracts of a puromycin-producing culture of Streptomyces alboniger which catalyzes the transfer of the methyl group from S-adenosyl-l-methionine-methyl-14C to O-demethylpuromycin. Propperties of this system are presented. The product has been characterized as puromycin, and the labeled methyl group has been shown to be located solely in the O-methyl-l-tyrosine moiety. A convenient synthesis of O-demethylpuromycin is described.
