Polyoma virus and SV40 are the smallest known tumor viruses. Their genomes have molecular weights of approximately 3.4 × 106 daltons, sufficient to code for approximately 200,000 daltons of protein. They are able to cause tumors in animals and alter the growth properties of infected tissue culture cells. Genetic analysis of these viruses is aimed at identifying the products of each of the viral genes and defining the mechanisms by which they alter the growth properties of infected cells. This paper describes some of the properties of temperature-sensitive mutants of polyoma.
