To investigate the association between elevated fetal hemoglobin (HbF) levels (≥1.5%) and non-deletional mutations within the promoter regions of the Gγ (HBG2) and Aγ (HBG1) globin genes.
Screened DNA samples from N=62 individuals presenting with fetal hemoglobin levels of 1.5% or higher.
Analyzed non-deletional sequence variations within the 5'-untranslated regions and promoter regions of both the HBG2 and HBG1 globin genes.
Identified four promoter variations in HBG2 (-158C/T, -309A/G, -369C/G, -567T/G) and four variations in HBG1 (+25G/A, -369G/C, -499T/A, -588G/A), alongside a homozygous -222/-225 AGCA deletion in HBG1.
High HbF levels correlated significantly with HBG2 promoter variants (-309A/T and -369C/G) as well as HBG1 promoter variants (+25G/A and -499T/A), suggesting a co-inherited haplotype effect.
