Homogeneous preparations of the potassium-requiring yeast aldehyde dehydrogenase are active with either DPN or TPN in the oxidation of a wide variety of aldehydes. Exogenous mercaptan is not a requirement for catalytic activity. The enzyme is denatured in 6 m guanidine hydrochloride to form a protein species with a molecular weight somewhat greater than 21,000. The denatured protein may be reassociated to yield functional enzyme of higher molecular weight.
