To elucidate the mechanism by which constitutively active Gα13 and receptor-mediated signaling regulate RhoA activation, invasiveness, and metastatic potential in melanoma.
Expressed constitutively active Gα13QL or applied lysophosphatidylcholine (LPC) to stimulate Gα13-coupled receptors in human melanoma cells.
Assessed RhoA activity, stress fiber assembly, cell invasion, and migration velocity following p190RhoGAP knockdown or co-expression of active RhoA.
Evaluated in vivo lung metastasis formation and overall survival using SCID mouse xenograft models.
Expression of Gα13QL or LPC stimulation activated p190RhoGAP, suppressing chemokine-induced RhoA activation, stress fiber assembly, and cell invasion.
Depletion of p190RhoGAP or introduction of constitutively active RhoA restored RhoA activation and rescued cell invasion defects.
