Alcian dyes may be used successfully to differentiate and locate sulfated and non-sulfated acid polysaccharides in algal tissues. Sulfated polysaccharides occur in Cladophora, Ulva, Taonia, Macrocystis, Pelagophycus, Pelvetia, Gigartina, and Smithora. In Macrocystis and Pelagophycus they predominate in the cuticle, outer cortex, and mucilage ducts. Acid, non-sulfated polysaccharides occur in Nostoc, Macrocystis, Pelagophycus, Phyllospadix, and Zea. Alginic acid predominates in the intercellular spaces of the medulla, sieve tubes, and inner cortex of Macrocystis and Pelagophycus.
