The 1H and 13C NMR spectra of the antibiotic novobiocin have been fully assigned by a combination of homonuclear and heteronuclear two‐dimensional techniques. One‐dimensional NOE difference spectra provide evidence for multiple conformations of the carbohydrate moiety with respect to the coumarin group. Mobile parts of the molecule gave positive NOEs whereas all other parts gave negative NOEs.
