A procedure for the rapid determination of internal amino acid sequences from a purified protein relies on covalent attachment of the protein to derivatized PVDF membranes followed by cleavage with specific proteases or chemicals. By combining different cleavage protocols with various attachment chemistries, new NH2-termini can easily be generated in various portions of the protein. Sequencing of insoluble or protease-resistant proteins is facilitated by the resistance of PVDF membranes to organic solvents.
