Polyribosomes were prepared from isolated rat liver cells by homogenisation or Triton X‐100 lysis. Triton X‐100 lysis proved the more effective method of preparation of ribosomes and was therefore used to study polyribosome profiles obtained from isolated liver cells incubated in Krebs‐Ringer phosphate and Eagle's basal essential medium for various times. Polyribosome aggregation decreased throughout the three‐hour incubation period but cells in the basal essential medium maintained a more aggregated profile and higher rate of albumin synthesis than those in Krebs‐Ringer phosphate.
