The effect of different methods of injection and the form of column-detector connection on band spreading in liquid chromatography has been investigated. Suitable systems which reduce the band spreading to a minimum are described. Column forms which will be compact and at the same time retain a satisfactory efficiency after bending are examined, and a suitable column shape is suggested. Results obtained by monitoring the elution curve of salt bands through a water/firebrick or water/silica gel chromatographic system demonstrates how the HETP of a liquid column varies with support particle size and column diameter. HETP curves are also given for the system using normal octane as a stationary phase supported on silanised celite with equilibrated acetone-water as the mobile phase. Examples of the separation of high molecular weight free fatty acids are included.
