A radioimmunoassay is described for a new polypeptide isolated from chicken pancreas (APP), and the distribution of this peptide among the pancreases of other birds, mammals, reptiles and amphibians was studied. APP-like immunoreactivity was found in acid-alcohol extracts of the pancreases of all eight avian species that were tested and in extracts of turtle and alligator pancreas. No cross-reactivity was found in the mammalian or amphibian pancreatic extracts or with the purified homologous bovine polypeptide. The anti-chicken APP sera did not cross-react with solutions of gastrin, insulin or glucagon and no APP-like reactivity was found in acid-alcohol extracts of any other chicken organ. Thus, APP was localized in the pancreas but no information is yet available on its cellular localization. APP immunoreactivity was demonstrable in chicken plasma and the concentration varied with the prandial state of the birds. This information, together with biological data previously published, suggests that APP represents another pancreatic hormone. (Endocrinology93: 558, 1973).
