Abstract.Prior treatment of human subjects with phenobarbital for ten days increased the rate of disappearance of an intravenous dose of14C‐labelled diphenylhydantoin in 12 patients. The phenobarbital treatment did not change the amount of ultrafiltrable diphenylhydantoin, nor the volume of distribution of diphenylhydantoin, and it is therefore considered justifiable to assume that phenobarbital may increase the metabolism of diphenylhydantoin in humans. In two patients who were intoxicated with barbiturates the half‐life of diphenylhydantoin was estimated to 4.25 and 3.75 hours, respectively, which are very low compared to the normal value of 11.3 hours.
