Growth of Chinese hamster fibroblasts in culture medium containing sucrose at a concentration of 0·08 M (S-medium), either before or after plating and irradiation, reduced the extrapolation number of the survival curve without affecting the slope. The effect diminished as the time between trypsinisation and irradiation increased, and appeared to be due to osmotic damage to the cells on transfer between trypsin of normal tonicity and S-medium.
