FK-506 effects on rat ventricular K+ currents occur independently of calcineurin; leaves open relevance to human electrophysiology and clinical use.
The functional effects of FK-506 on K+ currents and Ca2+ transients in rat ventricular myocytes are independent of calcineurin inhibition.
duBell et al. (1998) studied this question. FK-506 vs. Cyclosporin A was evaluated on Inhibition of K+ currents (Ito and IK) and calcineurin activity. FK-506 inhibited calcineurin activity by 65% and IK by ~60%, but calcineurin inhibition was not responsible for its functional effects on K+ currents in rat ventricular myocytes.
