Scalar couplings across N−H⋅⋅⋅N and N−H⋅⋅⋅O=C hydrogen bonds have been observed for the first time in the NMR spectra of nucleic acids and proteins. This finally allows the important hydrogen-bonding patterns in biopolymer structures to be mapped directly—from the exact value of the coupling constants even the length of the hydrogen bond can be derived.
