Key result
ANGPT-2 rs2442598 A/A genotype is linked to ~419% higher diabetic retinopathy risk in T1DM.
Why the study?
The study evaluated the relationship between the rs2442598 polymorphism and diabetic retinopathy.
Is the rs2442598 polymorphism in the ANGPT-2 gene associated with diabetic retinopathy in patients with type 1 diabetes mellitus?
Case-Control (n=236)
No
Is the rs2442598 polymorphism in the ANGPT-2 gene associated with diabetic retinopathy in patients with type 1 diabetes mellitus?
Odds Ratio: 5.19 (95% CI 1.21–22.27)
Absolute Event Rate: 10.3% vs 2.3%
p-value: p=0.027
The rs2442598 A/A genotype in the ANGPT-2 gene is significantly associated with an increased risk of diabetic retinopathy in Brazilian patients with type 1 diabetes.
rs2442598 A/A genotype was associated with DR risk; hypothesis-generating and requires prospective validation before clinical use.
OBJECTIVE: rs2442598 polymorphism and DR. METHODS: rs2442598 (G/A) polymorphism was genotyped by real-time PCR using TaqMan MGB probes. RESULTS: Genotype distributions of this polymorphism were consistent with the Hardy-Weinberg equilibrium. The frequency of the rs2442598 A allele was higher in cases compared to controls (p = 0.011). Moreover, the A/A genotype was more frequent in cases than in controls (p = 0.017) and was associated with risk for DR after adjustments for duration of DM, HbA1c, triglycerides, estimated glomerular filtration rate, and hypertension (odds ratio [OR] = 5.19, 95% confidence interval [CI] 1.21-22.27). This association was maintained under recessive (OR = 4.78, 95% CI 1.14-19.99) and additive (OR = 6.861, 95% CI 1.45-32.38) inheritance models. CONCLUSION: rs2442598 A allele and risk for DR in T1DM patients from southern Brazil. Additional studies are necessary to replicate this association in other populations.

Dieter et al. (2021) conducted a case-control in Type 1 diabetes mellitus and diabetic retinopathy (n=236). ANGPT-2 rs2442598 A/A genotype vs. G/G and G/A genotypes was evaluated on Diabetic retinopathy (OR 5.19, 95% CI 1.21-22.27, p=0.027). The ANGPT-2 rs2442598 A/A genotype was significantly associated with an increased risk of diabetic retinopathy in patients with type 1 diabetes mellitus (OR 5.19).
