To determine whether green tea polyphenol (-)-epigallocatechin-3-gallate (EGCG) suppresses COX-2 expression and its associated regulatory signaling pathways during colon carcinogenesis.
Treated human colorectal cancer cell lines HT-29 and HCA-7 with (-)-epigallocatechin-3-gallate (EGCG).
Evaluated constitutive COX-2 mRNA and protein levels, ERK1/2 and Akt pathway signaling, NF-kappaB-mediated promoter activity, and 3'UTR-mediated mRNA decay.
EGCG significantly inhibited constitutive COX-2 mRNA and protein overexpression in colorectal cancer cell lines.
Treatment downregulated ERK1/2 and Akt signaling pathways and reduced COX-2 promoter activity through the inhibition of NF-kappaB activation.
EGCG promoted rapid COX-2 mRNA decay mediated through its 3' untranslated region.
