Novel anthraquinone derivatives exhibit potent preclinical anticancer activity and potential to overcome drug resistance.
Structural modifications of anthraquinones may yield more effective anticancer drugs capable of overcoming drug resistance.
Anthraquinones show preclinical anticancer promise; leaves open clinical translation and resistance strategies pending trials.
Objective: In this study, some scientific studies on anthraquinone compounds that have been synthesized in recent years and have been found to have high in vitro and in vivo anticancer activity are summarized. It is aimed to contribute to the design and development of new anthraquinone derivative drugs that are more effective in cancer treatment and can overcome drug resistance.Result and Discussion: Anthraquinones are a privileged class of compounds that can be obtained from natural and synthetic sources and have anticancer properties with versatile mechanisms. Anthracyclines, which are potent topoisomerase inhibitors, are members of this class that are frequently used in cancer chemotherapy. Literature data suggest that new anthraquinone derivatives developed through structural modifications can exhibit better antiproliferative activity against resistant cancer cells than currently existing anthracyclines and may be more effective antitumor drug candidates.

Karayavuz et al. (2026) conducted a review in Cancer. Anthraquinones and their derivatives was evaluated. Newly synthesized anthraquinone derivatives, particularly those fused with heterocyclic rings, exhibit potent in vitro and in vivo anticancer activity and have the potential to overcome drug resistance.
