Strobel et al. (2019) studied this question. Frozen high-density HEK-293 cell stocks and CELLdiscs vs. Conventionally cultured cells was evaluated on AAV productivity. Frozen high-density HEK-293 cell stocks seeded directly into CELLdiscs demonstrated equal AAV productivity compared to conventionally cultured cells, yielding up to 1.2 × 10^15 vector genomes in 4 weeks.
