The VTTRL motif of TRPC5 is not necessary for activation but mediates the modulatory effect of EBP50 on channel activation kinetics.
Obukhov et al. (2004) studied this question. Co-expression of EBP50 with rTRPC5 vs. rTRPC5 without EBP50 or VTTRL-deletion mutant was evaluated on Activation kinetics (time-to-peak of histamine-evoked transient large inward current). Co-expression of EBP50 with rTRPC5 caused a significant delay in the time-to-peak of the histamine-evoked inward current, an effect dependent on the VTTRL motif.
