Gata4-induced PrE specification in mESCs drives ~81% blastoid formation efficiency with gastrulation potential.
p-value: p=≤0.0001
The study describes a high-efficiency method for generating blastoids with gastrulation potential using Gata4-induced PrE specification in mESCs.
Current blastoid protocols need refinement to resolve transcriptional ambiguity; leaves open whether factor-free methods can improve embryonic modeling fidelity.
Blastoids are regarded as promising models for further in vivo embryonic development 1 , 2 , 3 , 4 . Current blastoid protocols, which are based on either co-culturing embryonic stem cells/trophoblast stem cells (ESCs/TSCs) 2 , 3 or utilizing totipotency-like cells 1 , 5 , 6 , rely critically on exogenous factors such as FGF4, BMP4 and CHIR99021 1 , 2 , 3 , 5 , 6 to drive lineage specification and enhance blastoid formation. However, these blastoids harbor transcriptionally ambiguous cell populations and are insufficient for primitive endoderm (PrE) specification, and mostly arrest at pre-gastrulation stages 2 , 3 , 5 , 6 , 7 .

Guo et al. (2025) studied this question. Gata4-induced PrE specification in mESCs (iGata4-ESCs) vs. No doxycycline induction was evaluated on Blastoid formation efficiency (p=≤0.0001). Gata4-induced PrE specification in mESCs achieved highly efficient blastoid formation (up to 80.9%) and autonomous lineage segregation capable of developing to the gastrulation stage.
