Aortic 5-HT injection induces paralysis mimicking aortic embolism, whereas ligation or histamine alone fails to.
Why the study?
The relationship between aortic embolism and posterior paralysis in cats, and the role of thrombus-derived substances like 5-HT, was not fully understood.
May implicate 5-HT release from thrombi in feline aortic embolism paralysis; leaves open targeted serotonin blockade as hypothesis-generating.
The syndrome of naturally occurring aortic embolism in the cat is described. Reference is made to the experimental work of Imhoff who found that ligation of the aorta did not affect locomotion and that paralysis depended on the presence of a blood clot in the aorta. The production of histamine and 5‐hydroxy tryptamine (5‐HT) by the platelets of a thrombus is then mentioned and the literature on the pharmacology of 5‐HT is reviewed, the most important effect being the contraction of the smooth muscle of blood vessels. The author carried out experiments on forty‐nine cats: he confirmed Imhoff's assertion that neither single nor double aortic ligation caused paralysis; injection of histamine into the space between the double ligatures caused no effect but when 5‐HT was injected a syndrome similar to naturally occurring aortic embolism was produced.

HUGH C. BUTLER (1971) studied Aortic embolism and posterior paralysis (n=49). Injection of 5-hydroxy tryptamine (5-HT) between double aortic ligatures vs. Injection of histamine or aortic ligation alone was evaluated on Posterior paralysis / syndrome similar to naturally occurring aortic embolism. Injection of 5-HT between double aortic ligatures in cats produced a syndrome similar to naturally occurring aortic embolism, whereas histamine injection or ligation alone caused no paralysis.
