Key result
Betamethasone reduces clearance by ~36% and persists longer compared with dexamethasone.
Why the study?
Pharmacokinetic data for different dosage forms and routes of dexamethasone and betamethasone in Indian women were analyzed to characterize absorption and disposition parameters.
Population
48 healthy nonpregnant Indian women
Comparison
Single doses of 6 mg dexamethasone phosphate, betamethasone phosphate, or 1:1 betamethasone phosphate and acetate orally or intramuscularly
Design
Partial and complex cross-over pharmacokinetic study with two-compartment modeling

Betamethasone has slower clearance, larger volume of distribution, and longer half-life compared to dexamethasone, which may inform dosing when substituting these corticosteroids.
Krzyżański et al. (2021) conducted an RCT in Healthy nonpregnant women (n=48). Dexamethasone and Betamethasone vs. Cross-over comparisons between formulations and routes was evaluated on Population pharmacokinetic parameters (Clearance, Volume of distribution, absorption constants). Betamethasone exhibited slower clearance (5.95 vs 9.29 L/h), larger volume of distribution (72.4 vs 56.4 L), faster absorption, and longer persistence than dexamethasone.
