Key result
SC-52458 reduces the diastolic blood pressure response to Angiotensin II by ~91%.
Why the study?
The potency, efficacy, and tolerability of the new orally active AT1-receptor antagonist SC-52458 in normal volunteers were assessed to characterize its antihypertensive potential.
RCT (n=16)
Single-blind
Randomized
Effect estimate: decreased to 8.3% of baseline response
Absolute Event Rate: 2.6% vs 30.3%

SC-52458 demonstrates rapid, potent AT1 blockade in healthy volunteers; leaves open efficacy, dosing, and safety in hypertensive patients.
Hagmann et al. (1997) conducted an RCT in Healthy volunteers (n=16). SC-52458 vs. Placebo was evaluated on Diastolic blood pressure response to Angiotensin II challenges at 1 hour post-dose (200 mg) (decreased to 8.3% of baseline response). In healthy men, a 200-mg dose of the novel AT1-receptor antagonist SC-52458 decreased the diastolic blood pressure response to Angiotensin II challenges from a baseline of 30.3 mm Hg to 2.6 mm Hg at 1 hour post-dose.
