The current research work was intended to develop, validate and evaluate the environmental sustainability of an easy, rapid, accurate and eco-friendly reversed-phase high-performance thin-layer chromatographic procedure for the simultaneous measurement of AntiHIV agents Doravirine and Islatravir in pharmaceutical dosage form and bulk drugs. RP-18 F254S HPTLC plates were used for chromatographic separation and ethanol: water (70:30 v/v) proved to be the ideal mobile phase. Densitometric detection at 254 nm produced well resolved peaks with Rf values of 0.69 ± 0.03 for islatravir and 0.43 ± 0.02 for doravirine. The method was validated for linearity, specificity precision, accuracy, limit of quantification, robustness, limit of detection, and system suitability as per the ICH Q2(R2) guidelines. The technique exhibited good linearity for both the drugs within the concentration range of 200–1200 ng/band with acceptable correlation coefficients. This method was successfully applied for the assay of pharmaceutical dosage forms giving test values of 99.34% for Doravirine and 98.80% for Islatravir. The green performance of the analytical method was assessed using the Analytical Eco-Scale, AGREE, AGREEprep, Whiteness index and Complex Green Analytical Procedure Index (Complex GAPI) was confirmed by an Eco-Scale score of 87, an AGREE score of 0.79, AGREEprep score 0.76, Whiteness index score 80.0 (valid) and a predominantly green and yellow ComplexGAPI pictogram. The new sustainable RP-HPTLC method is reliable, accurate, simple, precise, economic and environmentally friendly and suitable for the routine quality control analysis of Doravirine and Islatravir in dosage form.
