A technique has been developed for observing the X-ray pattern of a protein crystal while its water content is altered by known and controlled changes in the vapour pressure of the atmosphere surrounding it.The method adopted is to equilibrate the crystal with the vapour above calcium chloride solutions of known concentration in an evacuated, thermostaticaUy-controlled enclosure. When crystals of horse met-haemoglobin are shrunk by this technique the lattice constants always change in a discontinuous manner, at a series of weU-defined vapour pressures.During this work five new types of haemoglobin lattice were observed.
